Some houses, like Stark and Greyjoy, have a chance of holding against a two-house assault, but will not win the game like this. The best they can do is hold the line, at which point hopefully they can convince one of the enemies it isn't worth attacking them. To make any progress, you will HAVE to make alliances even with the most defensive house.
So to the OP, the answer really is if all three neighboring houses ally against you, you're in deep already. Do your best to ally with Baratheon or Tyrell early on and get some traction. Remember AGoT is as much a sneaky-bastards game as much as a strategy game.
