Yes, a hero can use 2 shields to block a single attack.

A good tactic is to equip 2 shields whenever you are certain you won't need to attack before your next turn.

If possible, never sell a shield; they're good protection whenever you need to run somewhere.

The idea behind Weapon Mastery is that both hands must wield a weapon for it to work. It is thematic I guess. Wielding 2 one-handed melee weapons does nothing except allowing you a choice and making Weapon Mastery work if your best melee weapon is one-handed.
